The newly established research group „Targeted Oncology" in the Center for Oncology and Institute for Tumorbiology (group leader: Sonja Loges, M.D., Ph.D.) of the University Hospital Eppendorf in Hamburg, Germany is seeking a highly motivated

Post-Doc

Funding is limited to two years, but prolongation for another two years is expected. The project aims at unraveling mechanisms of resistance to molecular targeted therapies in preclinical mouse models of cancer and translation to clinical samples. The „Targeted Oncology" group is a young group within an internationally staffed institution with many internationel collaborators.

Your Project: Inhibiting angiogenesis to treat cancer was sucessfully translated from bench to bedside and prolongs life of many cancer patients worldwide. However, several open questions and drawbacks urge researchers back to the bench. First, efficacy is limited due to different mechanisms of resistance including upregulation of angiogenic rescue programs (clinical & preclinical studies), a systemic pro-inflammatory response (clinical & preclinical studies) and enhanced metastasis (clinical studies) among others. The projects aims at identification of novel targets at the tumor-host interface with a focus on cross-talk between tumor and bone marrow with the aim to minimize resistance towards anti-angiogenic drugs. Targets will be validated in state-of-the-art mouse models of cancer and in clinical samples using tissue microarrays. In addition, disseminated tumor cells will be studied to investigate impact of anti-angiogenic treatment on metastasis. You are expected to use a wide variety of in vivo and in vitro techniques to conduct these experiments. You will be culturing different tumor cell lines, implant them into mice and treat animals with different anti-angiogenic therapies. Subsequently, different cell populations will be FACS-sorted and subjected to Affymetrix gene arrays. Inhibition of promising targets will be studied mechanistically in combination with anti-angiogenic therapies and will be validated on human TMAs. You will be using FACS analysis, molecular biology, protein biochemistry and imaging for mechanistic analysis of impact on treatments on tumor biology. You will be performing RNA and protein isolation, ELISA, Western Blotting and QRT-PCR. You will transfect eukaryotic cells, use immunofluorescent and other techniques to stain mouse tissues and use confocal microscopy.

Your Skills: You sucessfully completed your PhD in biology, biochemistry, molecular biology or similar studies with publishing in peer-reviewed journals. You have experience in the described in vivo and in vitro techniques and are used to work independently. Knowledge in conducting mouse tumor models, molecular biology, bioinformatics (statistical analysis of gene expression data), protein biochemistry, confocal microscopy and histology are an advantage. Your written and spoken English is fluent. Motivation, flexibility and team spirit are your characteristics.
