The Fesik lab is a multi-disciplinary group in the Department of Biochemistry at Vanderbilt University, dedicated to oncology drug discovery. We are growing our cell biology group and have two opportunities for talented, motivated cell or cancer biologists to join this dynamic group: one research associate and one postdoctoral associate. Both scientists will work closely with students, post-docs, biologists, and chemists to screen compounds in cell-based assays, develop new assays and techniques, and contribute to the discovery and validation of new targets for cancer drug discovery.
Key Functions (dependent upon level and training):
• Conduct cell-based and plate-based assays to enable drug discovery efforts. Use established protocols and existing assays to measure growth inhibition, apoptosis, regulation of transcription, and inhibition of signaling pathways; assays include fluorescence- and luminescence-based formats, such as ELISA, In-cell Western, and FRET. Screen compounds in single concentration and dose-response schemes, analyze and report data.
• Contribute to the design and development of novel assays to measure the functional consequences of inhibition of a number of diverse targets, including assays to measure disruption of protein-protein interactions, anchorage-independent growth, migration, invasion, and signaling.
• Participate in research projects to identify and validate new targets for cancer drug discovery. Experiments will include target silencing by RNAi, target overexpression, and compound treatment of cancer cell lines.
• Conduct research projects to understand the mechanism of action of novel targeted therapies identified in our group. This may include screening cancer cell lines and tumors to understand the genetic predictors of response to these targeted agents.
• Perform additional lab duties, such as preparing buffers and reagents, ordering supplies, and maintaining equipment.
Minimum Requirements:
The research associate position requires a Bachelors or Masters degree in cellular, molecular, or cancer biology, or a related field and a minimum of 3 years laboratory experience. The candidate must be well-versed in mammalian cell culture, including passaging and storage of cell lines, transfection protocols, and harvesting and preparing cell lysates. Working knowledge of plate-based assays, Western blotting techniques, PCR, and cloning and expression protocols are required. Excellent data analysis capabilities and proficiency with curve-fitting and graphing programs are essential. Applicants with previous compound screening or drug discovery experience in a pharmaceutical or biotech company will be given special consideration.
The postdoctoral candidate should have a Ph.D. in cell or cancer biology or a related field and be interested in receiving additional training in the discovery and pre-clinical development of cancer therapeutics.
