Description:

In a Wilhelm-Sander Stiftung funded project we aim at utilization of Bone morphogenetic proteins as novel therapeutic agents for future treatment of adrenocortical carcinomas.

Adrenal masses are one of the most common endocrine tumors diagnosed. Although most adrenal tumors are endocrine inactive adenomas, a considerable proportion is associated with hormonal hyperfunction and/or malignancy. Despite constant clinical improvements the overall prognosis of patients with adrenocortical carcinoma (ACC) is still poor and new therapies are urgently needed. Through detailed functional investigations we could recently demonstrate a modulating effect of Bone morphogenetic proteins (BMP) on adrenocortical tumor growth and steroidogenesis (Johnsen et al. Cancer Res. 2009 Jul 15;69(14):5784-92). Moreover BMP2 and 5 are lost or down-regulated in a large proportion of ACC samples. The general aim of this proposal is to utilize the results from our in vitro and in vivo studies as a starting point to develop novel BMP based strategies with therapeutic potential for adrenocortical cancer. This aim will be targeted in a multi-step approach by applying methods that either directly or indirectly restore BMP levels in adrenocortical cancer cells. Molecular and functional impact of these interventions will be investigated in appropriate in vivo models.

At the center of Endocrine Research of the Medical University Clinic Innenstadt (www.endocrine-research.mki.klinikum.uni-muenchen.de) a young and highly motivated team of basic and clinical scientists is working on a number of aspects of endocrine tumors and hormonal autonomy. Through regular lab seminars and close scientific mentoring within the team a successful completion of the project will be achieved. The group has an internationally renowned expertise in the field of adrenal tumorigenesis with a successful track record in publication and funding.
